1. Histology and immunohistochemical staining

SERVICES OFFERED Services provided by this component of the Core include routine histology (tissue trimming, embedding, sectioning, H&E staining, histochemical staining and immunohistochemical staining) and the retrieval of paraffin-embedded blocks of normal and diseased tissue (by diagnosis) from our archives. A detailed list is provided on the order form.
COMPLIANCE IRB or IACUC approval is required for all services requiring the use of human or animal tissue, respectively. Service requests will NOT be processed without the necessary compliance information (provide copy of appropriate approval form to Dr. Carpenter; also see COMPLIANCE guidelines).
COST TO INVESTIGATOR Re-charge rates are listed on the order form.
ORDERING SERVICES Select either the PDF or WORD version of the Histology service order form, complete the form and fax (714-456-5873) or email to Ms. Farah Akhtar.

Investigators may use our departmental courier service to send samples to UCIMC, where the Histology unit is located. Processed samples can also be returned by courier. To send samples via courier, email the request form to Ms. Akhtar (specifying that samples will follow via courier) and include a copy of the request form with the samples. Pack the samples securely in a sturdy box. Tightly seal any containers filled with formalin. Address the box to Pathology Research Services Core, c/o of Ms. Farah Akhtar, Bldg. 10, UCIMC.

Be sure to specify that samples are to be returned by courier also. Take the box to the courier pickup location in the Hitachi Bldg. (Room 214) on the School of Medicine campus. Mr. Sutherland will send you an email on the day that samples are sent back to the Hitachi Bldg. drop by return courier.

2. Morphometry and image analysis of tissue sections

SERVICES OFFERED This component of the Core provides services required to quantify cellular elements or molecular signals detectable by microscopic evaluation of tissue sections. This includes determining the number and/or size of cellular organelles; measuring the intensity of immunostaining of individual cells in a tissue section, and counting the percentage of cells and/or nuclei that are immunostained with a particular antibody. These analyses can be performed on sections which were H&E stained or immunostained in the Histology component of the Core, or on sections prepared by individual investigators.

3. Laser-capture micro-dissection of tissue sections

SERVICES OFFERED This component of the Core allows investigators to fractionate cell populations from tissue sections. For example, normal tissue can be purified from tumor tissue on tissue sections containing a mixture of these cell types. The core facility, located in Hewitt Hall, contains a Leica CM3050 cryostat and a Leica LS-AMD outfitted with fluorescent optics (allowing the selective visualization and capture of immunostained cells), with a minimum beam width of less than 1 µm.

This service can be performed on sections that were H&E stained in the Histology component of the Core, or on sections prepared by individual investigators.

COMPLIANCE IRB or IACUC approval is required for all services requiring the use of human or animal tissue, respectively. COST TO INVESTIGATOR All users must first undergo a two hour orientation on the instrument with Dr. Edwards. There is no fee for UCI investigators, and $100 per hour for non-UCI affiliated users. All investigators requiring the services of a surgical pathologist to perform the microdissection will be charged $100 per hour of LCM time. Investigators must purchase appropriate caps with laser-capture membrane (Arcturus), or foiled-coated slides (Leica).
ORDERING SERVICES Contact Dr. Edwards to arrange services. Users performing their own microdissections must receive training on the use of the LCM from Dr. Edwards. LCM time must be reserved in advance and each session must be recorded in the log book.
   
4. Analyte Determination (Clinical Pathology Services)

SERVICES OFFERED This component of the Core provides investigators at UCI, other medical centers, and industry with the complete spectrum of clinical pathology (laboratory medicine) services that are available for patient care at UCIMC. COMPLIANCE IRB approval is required for all services performed as part of a clinical research study at UCI. Service requests will NOT be processed without the necessary compliance information (provide copy of appropriate approval form to Dr. Emerson).
COST TO INVESTIGATOR Re-charge rates are determined on a case-by-case basis in consultation with Dr. Emerson or the UCI Clinical Research Finance Administration (for specimens processed as part of a UCI clinical research study). 5. Tissue bank and procurement service (TBPS)

SERVICES OFFERED The Core will bank remnants of tissue (both normal and tumor) resected from patients undergoing surgery at UCIMC. Remnant tissue is the residual portion of a surgical specimen following processing for diagnostic purposes (see Tissue Custody Policy). Signed consent from each patient will also allow us to obtain peripheral blood samples (serum and DNA) and clinical follow-up at 6 month intervals for 3 years. Investigators may request access to tissue and blood samples, as well as clinical data. Requests for large amounts of tissue or multiple samples, or access to relatively rare specimens will be reviewed by the Core’s Tissue Use Committee.
COMPLIANCE IRB approval is required for access to this tissue. Service requests will NOT be processed without the necessary compliance information (see order form and COMPLIANCE guidelines; provide copy of appropriate approval form to Dr. Narula). Investigators are encouraged to contact Dr. Narula concerning the availability of tissue prior to submitting projects for IRB approval.

6. Genotyping (includes nucleic acid extraction from blood and tissue sections)

SERVICES OFFERED Genotyping services are performed in the Diagnostic Molecular Pathology lab and include a variety of PCR and/or Southern blot based analyses of mutant or polymorphic alleles. The lab will work with investigators to identify genotype-based tests that may be of value in clinical treatment and cancer control studies. This service can also perform mRNA-based testing for occult cancer or residual disease. In addition, this component of the Core provides automated nucleic acid extraction from whole blood, paraffin-embedded or frozen tissue sections.

COST TO INVESTIGATOR Nucleic acid extraction re-charge rates are listed on the order form. Re-charge rates are determined on a case-by-case basis in consultation with Dr. Krolewski or the UCI Clinical Research Finance Administration (for specimens processed as part of a UCI clinical research study).
